Manganiello applied to the Carnegie Mellon School of Drama after graduating from high school, but he was not accepted. He then decided to enroll at Pittsburgh University. Joe Manganiello made another attempt to get into Carnegie Mellon School of Drama a year later, and this time he was accepted. Manganiello earned a Bachelor of Fine Arts in acting in 2000. Manganiello moved to New York and Los Angeles after graduating in an attempt to launch a career in acting. He took part in several stage shows.

Sam Raimi’s “Spider-Man,” which debuted in 2002, marked Manganiello’s feature film debut. Following the release of this film, the actor was cast in other small roles in various television shows. He starred in episodes of Scrubs, Medium, E.R., and C.S.I. Manganiello played a recurring part in the television sitcom “How I Met Your Mother” from 2006 until 2012.
