In June 2008, Asamoah signed a contract with Serie A club Udinese, where he rapidly established himself as a vital member of the team and the country. Several elite European clubs started to show interest in him after he had strong results at the 2010 World Cup and Africa Cup of Nations. Under French manager Claude Le Roy, he made his debut for the Ghana national team, the Black Stars, in 2006.

He competed for Ghana in the 2012 and 2013 tournaments after winning a bronze and silver medal in the Africa Cup of Nations in 2008 and 2010, respectively. Additionally, he represented Ghana in the FIFA World Cups of 2010 and 2014.

He made his Supercoppa Italiana 2012 debut on August 11 against Napoli, where he scored a goal that gave Juventus the overtime victory over Napoli. Antonio Conte, the Juventus coach at the time, decided to start him in the opening league game of the 2012–2013 season against Parma after he had such an impressive performance against Napoli.

He had an instant effect on August 25, setting up Stephan Lichtsteiner’s goal in a victory at home. Juventus purchased Asamoah outright in June 2013. After that, he agreed to a four-year deal with the team. Asamoah scored a memorable goal in Juventus’ home victory over Florentina on March 9, 2014, and the team went on to successfully defend both the Serie A title and the Supercoppa Italiana that season.

He was sidelined for the majority of the season after suffering a serious knee injury in the league opener against Empoli on November 1, 2014.

Asamoah led Juventus onto the field in the opening match of the 2016 International Champions Cup Australian edition on July 23, 2016, following the clubs’ draw with Melbourne. Asamoah hobbled off the field the day before Palermo with a sprained right knee; tests at the J-Medical center revealed he had sustained a complete tear of the medial meniscus ligament. On September 25, 2016, it was confirmed that Asamoah would be out for six weeks. A report from January 2018 stated that Asamoah made the decision not to renew his contract with Juventus, which was set to expire in June of that same year.

He made his 150th appearance for Juventus in a Serie A road draw against S.P.A.L. on March 17. Asamoah committed to join Inter in May 2018, and his formal signing was finalized in July upon Lega Serie A’s registration in July 2018.
